2022年以来,我院动物及医学方向在Science Advances、Cell Research、PNAS、Molecular Therapy、Signal Transduction and Targeted Therapy等主流期刊上发表论文16篇,围绕着生殖、代谢及发育,基因组学与编辑,疾病发病机制及治疗等领域取得了一系列前沿成果。
生殖、代谢及发育领域:张华课题组揭示了哺乳动物成年卵巢血管新生的发育模式以及生理意义,提出了延缓女性生殖衰老的新策略(Xu et al., 2022);阐明了卵巢中卵泡发育多态性的形成机制,并揭示了胚胎性腺卵母细胞发育进程与青春期始动时间之间的相关性(Dai et al., 2022)。侯云鹏课题组发现微塑料暴露会引起小鼠卵巢炎症反应,并诱发一系列的卵泡发育异常及生殖障碍,为微塑料的生殖毒性机制提供了证据(Liu et al., 2022)。李向东课题组通过临床与基础研究结合,明确了miR-335-5p在多囊卵巢综合征发病过程中的作用,相关研究成果提示了miR-335-5p作为PCOS潜在诊断生物标志物和治疗靶点的可能性(Zhang et al., 2022);同时,李向东课题组解析了Adtrp在调控产热脂肪组织发育及代谢的分子机制,为动物基础代谢的调控机制提供了新的视角(Li et al., 2022)。于舒洋课题组围绕免疫细胞发育,揭示了SRSF1通过调控Runx3的表达,调控CD8+细胞的晚期发育,为理解T细胞晚期发育和谱系选择的转录后层级调控提供了新的思路(Ji et al., 2022)。
基因组学与编辑领域:胡晓湘课题组与复旦大学舒易来教授课题组合作使用CRISPR/CasRx (RfxCas13d) 基因编辑系统在体内治疗遗传性耳聋 (Zheng et al., 2022),为其在听力及其他许多其他遗传性疾病中的进一步应用铺平了道路。赵毅强课题组基于单倍型量化大花白猪基因组量中自北方大型猪的渗入片段提出了两步渗入的群体历史模型,确定了IGF1R、SRC和PCM1等可能影响体型的候选基因(Wang et al., 2022);并分析了线粒体异质性在四代家系中的动态变化,揭示了线粒体变异近中性的传递规律(Liu et al., 2022)。
疾病发病机制及治疗领域:于政权课题组发现了一种特殊的能够响应饮食状态变化的瘦素受体阳性(Lepr+)间充质细胞,为制定肠道相关疾病的营养干预策略提供重要的科学理论指导(Deng et al., 2022)。李向东课题组揭示了一种新的SIRT1调控蛋白-NOC4L,在核仁胁迫下调控p53的分子机制(Jia et al., 2022),为深入了解DNA损伤早期的肿瘤发生和细胞自我保护机制提供了依据;同时,该课题组通过对三个肝细胞癌(HCC)的RNA-seq数据库集成分析并结合临床病人样本, 揭示了环状RNA导致化疗耐药的新机制(Li et al., 2022)。英郑欣课题组同北京生命科学研究所王晓东课题组合作发现MLKL在糖尿病神经病变的小鼠模型中介导髓鞘降解,同时证实该机制存在于糖尿病神经病变的病人中,而MLKL小分子抑制剂可有效保护糖尿病神经病变小鼠神经的结构与功能(Guo et al., 2022)。陈忠周课题组先后解析了人去甲基化酶家族成员ALKBH6及与辅因子的复合物、人副流感病毒3的N-P蛋白复合物、人畜共患病毒裂谷热病毒L蛋白的晶体结构,明确了ALKBH6优先结合的核酸类型及与抑癌因子ZMYND11互作关系,阐明N蛋白构象变化的机制和P蛋白的伴侣机制,为相关分子的机制研究奠定重要的基础 (Ma et al., 2022; Dong et al., 2022l; Wang et al., 2022)。
